เทศบาลเมืองสองพี่น้องตั้งอยู่ในเขตตำบลสองพี่น้อง อำเภอสองพี่น้อง จังหวัดสุพรรณบุรี มีพื้นที่ 10.4 ตารางกิโลเมตร (6,500 ไร่) อยู่ห่างจากศาลากลางจังหวัดสุพรรณบุรีประมาณ 42 กิโลเมตร และอยู่ห่างจากกรุงเทพมหานครประมาณ 104 กิโลเมตร

ทิศเหนือ ติดต่อกับเขตตำบลบางพลับและตำบลศรีสำราญ
ทิศใต้ ติดต่อกับเขตตำบลเนินพระปรางค์และตำบลบางเลน

2 ลักษณะภูมิประเทศ
เทศบาลเมืองสองพี่น้องมีพื้นที่ราบลุ่มเป็นส่วนใหญ่ เหมาะสำหรับการทำการเกษตร มีลำคอลง หนอง บึงอยู่ทั่วไป และคลองสองพี่น้องเป็นสายสำคัญ ในฤดูฝนจะเกิดน้ำท่วมในบางชุมชน เช่น ชุมชนไผ่หมู่ ชุมชนสองพี่น้อง ชุมชนบางใหญ่
3 ลักษณะภูมิอากาศ
สภาพอากาศโดยทั่วไปมีลักษณะ ร้อนชื้น
ฤดูร้อน ได้รับอิทธิพลจากลมตะวันออกเฉียงเหนือและลมฝ่ายใต้พัดผ่าน ตั้งแต่กลางเดือนกุมภาพันธ์ ถึง กลางเดือนพฤษภาคม
ฤดูฝน ได้รับอิทธิพลจากมรสุมตะวันตกเฉียงใต้ ตั้งแต่กลางเดือนพฤษภาคม ถึง กลางเดือนตุลาคม
ฤดูหนาว ได้รับอิทธิพลจากมรสุมตะวันตกเฉียงเหนือ ตั้งแต่กลางเดือนตุลาคม ถึง กลางเดือน

4 สภาพทางสังคม
1 การศึกษา
เทศบาลเมืองสองพี่น้องให้บริการด้านการศึกษาตั้งแต่ระดับอนุบาลจนถึงระดับมัธยมศึกษาตอนต้น เทศบาลได้ตระหนักถึงความสำคัญของการพัฒนาเด็กในช่วงอายุ 2 ½ -3 ปี ซึ่งเป็นวัยสำคัญของการวางรากฐานคุณภาพชีวิต จึงได้มีการจัดตั้งศูนย์พัฒนาเด็กเล็ก และได้ดำเนินการก่อสร้างอาคารศูนย์พัฒนาเด็กเล็กและอาคารประกอบ ในปีงบประมาณ พ.ศ. 2552 ในเขตเทศบาลยังมีโรงเรียนในสังกัดของกระทรวงศึกษาธิการ 1 โรงเรียน และมีโรงเรียนเอกชน 1 โรงเรียน เพื่อให้บริการด้านการศึกษาแก่เยาวชนในเขตเทศบาลและพื้นที่ใกล้เคียง ประกอบด้วย

2 สาธารณสุข
ด้านการสาธารณสุขเทศบาลเมืองสองพี่น้องได้ให้บริการด้านการดูแลสุขภาพแก่ประชาชนในเขตเทศบาล โดยมีโรงพยาบาลส่งเสริมสุขภาพ จำนวน 1 แห่ง ศูนย์บริการสาธารณสุขชุมชน จำนวน 1 แห่ง มีโรงพยาบาลของกระทรวงสาธารณสุข 1 แห่ง คือ โรงพยาบาลสมเด็จพระสังฆราชองค์ที่ 17 และมีคลินิกเอกชน จำนวน 9 แห่ง เทศบาลเมืองสองพี่น้องได้ร่วมมือกับโรงพยาบาลสมเด็จพระสังฆราชองค์ที่ 17 ในการจัดทำโครงการศูนย์สุขภาพชุมชนบางลี่ ซึ่งทางโรงพยาบาลฯ จัดให้มีแพทย์มาประจำที่ศูนย์บริการสาธารณสุขเทศบาลเมืองสองพี่น้องทุกวันอังคาร พร้อมทั้งจัดตั้งศูนย์สาธารณสุขมูลฐาน จำนวน 19 ชุมชน โดยมีอาสาสมัครสาธารณสุขชุมชนในเขตเมือง (อสม.) ผู้ผ่านการอบรม เป็นผู้ให้ความรู้และคำแนะนำด้านการสาธารณสุขพื้นฐานแก่ประชาชนในชุมชน
3 อาชญากรรม
ในด้านความปลอดภัยในชีวิตและทรัพย์สินนั้น เทศบาลเมืองสองพี่น้อง อยู่ในพื้นที่รับผิดชอบของสถานีตำรวจภูธรสองพี่น้อง และเทศบาลในฐานะองค์กรปกครองส่วนท้องถิ่นซึ่งมีหน้าที่ในการรักษาความสงบเรียบร้อย การส่งเสริมและการสนับสนุนการป้องกันและรักษาความปลอดภัยในชีวิตและทรัพย์สิน รวมทั้งการป้องกันและบรรเทาสาธารณภัยต่างๆ จึงเป็นหน้าที่ความรับผิดชอบของเทศบาลเมืองสองพี่น้อง ซึ่งมีงานป้องกันและบรรเทาสาธารณภัย ทำหน้าที่ตรวจตรา ดูแล และรับผิดชอบ โดยมีการจัดตั้งอาสาสมัครป้องกันและบรรเทาสาธารณภัย เพื่อเป็นความร่วมมือระหว่างประชาชนในชุมชนกับท้องถิ่น
4 ยาเสพติด
เทศบาลเมืองสองพี่น้อง ได้ดำเนินโครงการตามอำนาจหน้าที่ และเป็นไปตามยุทธศาสตร์การพัฒนาของเทศบาลเมืองสองพี่น้องเพื่อให้เกิดความปลอดภัยในชีวิตและทรัพย์สินของประชาชน โดยได้ดำเนินโครงการ ดังนี้
- โครงการติดตั้งกล้องโทรทัศน์วงจรปิด (CCTV) พื้นที่เสี่ยงภายในชุมชนเขตเทศบาล
- โครงการก่อสร้างลาน กีฬาต้านยาเสพติด เทศบาลเมืองสองพี่น้อง
